Apomorphine is a non-narcotic derivative of morphine that acts as a dopamine agonist without stimulating opioid receptors. Dopamine, nicknamed the "pleasure chemical", is commonly thought of as the main chemical of pleasure. However, dopamine actually regulates the brain's perception of the level of satisfaction, or anticipated satisfaction, certain activities or outcomes produce - which is experienced as the feeling of pleasure. This process of pleasure association dictates the level of desire we have to engage in certain activities, like sex. Dopamine also increases blood flow to the genitals by blocking norepinephrine release, leading to better erections in men and increased sensation in women. Although the primary actions of apomorphine are on dopamine receptors, it also has some weak affinity for seratonin receptors which can improve mood. When taken prior to sexual intercourse, apomorphine will increase the association of pleasure with sexual activities, improving satisfaction and desire (libido).
 
Oxytocin is a peptide hormone produced in the hypothalamus of both men and women. It is commonly called the "love hormone" or "cuddle drug" due to its roles in reward responses, decreased anxiety, social bonding, maternal behaviors, romance, arousal, sexual activity and reproduction, and orgasm and sexual satisfaction. Oxytocin increases drastically during sexual activity in both males and females, and is believed to be responsible for emotional bonding with sexual partners. It is also thought to decrease auditory noise from the environment to allow for better perception of emotional and social cues, which may improve feelings of connection and intimacy. The production and secretion of oxytocin is controlled via a positive feedback mechanism, meaning initial release stimulates further production and release. Bremelanotide (PT-141) is a peptide that acts as a non-selective agonist of melanocortin receptors, which control sexual function/arousal, appetite, immunity, and other physiologic actions. While it was originally studied as a sunless tanning agent, researchers quickly noticed that males achieved spontaneous erections and increased rates of ejaculation. Bremelanotide is currently only FDA-approved for treatment of Hypoactive Sexual Desire Disorder (low sexual desire) in premenopausal women, however, many studies have documented improved sexual outcomes in men as well. Stimulation of melanocortin receptors produces different mechanisms of action in males, increases nitric oxide in the penis leading to vasodilation and erection, compared to females, stimulates dopamine release in brain areas responsible for sexual motivation, arousal, and desire. Unlike phosphodiesterase inhibitors which act directly on vascular structures, bremelanotide increases neurotransmitter signaling to indirectly stimulate genital blood flow while increasing sexual desire responses in the brain. The Pleasure Enhancement pills utilize the different mechanism of action to amplify the individual effects and maximize sexual desire, arousal, and pleasure. 
 
Tadalafil is a PDE5-inhibitor that dilates blood vessels to increase blood flow to the penis and treat erectile dysfunction in men. Women do not typically rely on genital blood flow as much as men, but rather lubrication from sexual excitement. Likewise, tadalafil does not work in men unless they are sexually excited.
